TGF-beta and scarring: What's next in wound healing modulation?

In a recent phase-2 clinical trial, a TGF-b2-specific scavenging antibody was not superior to control in preventing filtering bleb scarring. Nevertheless, several data suggest TGF-b as a pivotal mediator of filtering bleb failure. The modulation of intracellular signals elicited by TGF-b using small molecule inhibitors is another strategy to prevent TGF-b-induced tissue alterations. Cell biological studies reveal the kinases p38 and ROCK as possible targets for such an approach.
