AbstractP 072 Retinotoxity of Amiodarone in mice Turdumambetova G., Bredehorn T., Claus J., Simon E., Duncker G. I. W. Department of Ophthalmology of the Martin-Luther-University Halle-Wittenberg Objective: Functional and morphological changes in mice retina after application of amiodarone. Methods: 20 pigmented mice of the stock Black CL 57 treated with amiodarone (150-200 mg/kg) for 36 weeks and 20 control mice were examinated during 70 weeks with ERG, funduscopy and electron microscopy. Results: The comparison between amiodarone- and control mice showed a few changes. The oscillatory potentials were passing changed in a 100 mcds/m2 light intensity and none in 3000 mcds/m2. Mildly appearing funduscopical changes were nearly reversible after the withdrawing of the treatment. In some mice founded vague granular inclusions were histological predominantly in the photoreceptor layer. They were also reversible till the end of the study. Conclusions: The present results suggest reversible mild retinotoxic effect of amiodarone in mice that could be proved by functional and morphological investigations.
